Lyophilised vs Liquid: Reconstitution, Storage and Getting the Dose Right
Powder or pre-mixed? The answer changes your cost, your shelf life and how many ways there are to make a mistake in week one.

The first vial is where most mistakes happen, and nearly all of them are the same three mistakes. None of them are complicated once someone explains the process properly, which is what this is.
What lyophilised means
Lyophilisation is freeze-drying. The peptide is frozen, then the surrounding water is removed by sublimation under vacuum — going straight from ice to vapour without passing through liquid. What is left is a dry cake or a thin film at the bottom of the vial.
Sometimes that cake is obvious. Sometimes, particularly at low milligram amounts, it looks like the vial is empty. It is not. A 5 mg vial contains 5 mg of a light powder, which is very little material spread across a wide glass base.
Why most peptides ship as powder
Peptides in solution degrade. Hydrolysis, oxidation and aggregation all proceed far faster in water than in a dry state. A lyophilised peptide stored correctly is stable for a long time; the same peptide in solution has a shelf life measured in weeks.
That is the entire trade-off. Powder buys stability and costs you a preparation step. Liquid buys convenience and costs you shelf life.
Reconstitution, step by step
- Let both vials reach room temperature. Injecting cold water into a cold vial increases the chance of the powder clumping.
- Swab both stoppers. The peptide vial and the bacteriostatic water vial, with a fresh alcohol swab each. Let them dry.
- Draw your bacteriostatic water. Use the volume your protocol specifies — this number determines your concentration, so it is not arbitrary.
- Inject it down the side wall of the vial. Not directly onto the powder. Angle the needle so the water runs down the glass. Peptides are fragile and a direct jet can shear them.
- Do not shake. Swirl gently, or set it down and wait. Shaking denatures peptides. It should go clear on its own within a few minutes.
- Refrigerate. Once reconstituted it lives in the fridge, not the door of the fridge, and not the freezer.
If the solution is cloudy, has visible particles, or has changed colour, do not use it.
The maths people get wrong
This is the one that causes real problems. Concentration is total peptide divided by total water added.
5 mg of peptide + 2 mL of bacteriostatic water = 2.5 mg per mL. On a standard 1 mL insulin syringe marked in 100 units, 1 mL = 100 units, so each unit carries 0.025 mg. A 250 mcg dose is therefore 10 units.
Two rules keep you safe. First, write the concentration on the vial label in marker the moment you mix it — future you will not remember. Second, if you are ever unsure of the arithmetic, ask before you inject, not after. Nobody has ever been annoyed by that question.

That 28-day figure on bacteriostatic water is the one people miss. The benzyl alcohol preservative is what makes multi-dose use possible, and its protective window is limited once the stopper has been pierced.
Light matters more than most people assume. Keep vials in the box they arrived in.
When to choose liquid
Choose the ready-to-use presentation if this is your first protocol, if you travel frequently, or if you simply do not want a preparation step in your week. There is no performance penalty — the compound is the same compound. You are paying for the pharmacy to have done the mixing under sterile conditions.
Choose lyophilised if you want the longest shelf life, the lowest cost per milligram, or the ability to set your own concentration.
A short checklist
- Room temperature before mixing
- Swab both stoppers, let them dry
- Water down the wall, never onto the powder
- Swirl, never shake
- Write the concentration on the vial
- Refrigerate, keep it dark, use a fresh syringe every time
